Founded in 2022 around the Yale School of Medicine research of scientific co-founders Stefan Somlo and Michael Caplan, it is led by co-founder and CEO Wayne Sackett and operates from Menlo Park, California and Newton, Massachusetts. The company launched from stealth in October 2026 with $103 million in committed financing to advance its AAV gene therapy candidates EST-201 and EST-101 toward the clinic.
